Step 1: Decide weither you have one or two USB cables joined together, it should look like a 4 PIN header.

Step 2: Get your manual from ECS: http://www.ecs.com.tw/ECSWorldWide/Index.aspx?MenuID=0&LanID=9

Step 3: Install the 4 PIN header the right way +5V, - DATA, + DATA, GND on the motherboard, refer to the manual if you get stuck.

I have a ECS A740GM-M Motherboard in an Asus TX-381 case. I can't get the 2 front usb ports to work. What am I not wiring up right?

Make sure you are putting the front usb header connector on the proper USB pin connection on the motherboard. You should see nine pins in 2 rows together. Check the manual that came with the motherboard to find the correct pins location.

Tower top mounted USB portss

The cables from the front panel USB connectors have a key in them that will line up with the missing pin on the board's USB headers. Just line up the connectors and plug them onto the board. It doesn't matter if you use USB1 on the board for the plug marked USB1.
