There are several things that cause misfires #1 spark plug #2 ignition coil and plug boot.#3 plugged fuel injector #4 engine cranking compression low #5 Engine running compression low #6 vacuum leak at that intake runner. Being that the newer computer systems can find misfires pretty easy suggest to do this remove # 2 plug and inspect gap and its outer shell for burn makes compare to another cylinder. all looks good then swap plug coil and boot to the other cylinder if miss follows to components then replace then if stays with cylinder this is much harder to fix.

do a compression test on the engine.if that checks out ok then the problem is either an ignition problem or a bad fuel injector

theres a possibility that the #2 fuel injector is clogged up. go to any auto parts store and tell them your problem, they should have a fix. unless the injector is bad. good-day!! are you sure #2 wire is ok! You probably need to clean out the MAF/sensor as well as throw in some injector cleaner.

Causes of p0300, p0302, p0304, p0306 in 2000 toyota avalon

P0300 = Random/multiple cylinder(s) - misfire detected.
P0302 = Cylinder 2 - misfire detected.
P0304 = Cylinder 4 - misfire detected.
P0306 = Cylinder 6 - misfire detected.

A misfire can be caused by a few things. The most common causes are usually spark supply or fuel supply. Get BOTH of these checked.
All the misfires are on the same side of the engine so I would suspect a fuel rail or an injector issue on that side of the engine.
